Course Goal

At the end of this course students will be able to explain prodrugs, enzymatic or non-enzymatic biotransformation events, enzymes that take role in biotransformation, steps of biotransformation, role of liver in biotransformation, change in activity of enzymes, effect of biotransformation on bioavailibility and clearance.

Course Content

Prodrug, Basic properties of biotransformation event and enzymes, Structures consists biotransformation enzymes, Non-enzymatic biotransformation, Types of enzymatic events, Oxidation, Cytochrome P450 enzymes, Oxidation events facilitated by cytochrome P450 enzymes, Epoxide formation, Oxidation events facilitated by non-microsomal enzymes, Reduction, KOPMA, Conjugation, Role of microsomal enzymes of liver on drug metabolism, Dependence of metabolism pattern and speed to species, Microsomal enzyme induction, Inhibition of microsomal enzymes by drugs, Enzyme activation, Hepatic clearance of drugs and change of clearance in hepatic diseases, Importance of ratio of drug excration with regard to bioavailibility, contribution of drug metabolism studies to drug development.
