Course Goal / Objective

Investigation of post-translational mechanisms involved in controlling protein function and fate in organisms at different organizational levels

Course Content

Introduction to post-translational modifications, definitions, and basic functions; types of post-translational modifications and their roles in cell signaling; phosphorylation types and their effects on protein functions; ADP-ribosylation and its effects on protein functions; methylation types and their effects on protein functions; acetylation (Nα- and Nε-acetylation) types and their effects on protein functions; glycosylation and the role of sugars in protein functions; lipidation and the role of lipids in protein functions; S-nitrosylation, S-sulfidration, S-glutathionylation, and their mechanisms; adenylation, uridylation, and methionine oxidation; ubiquitination and its mechanisms; ubiquitin-like modifications and their mechanisms; specific polypeptide cleavage mechanisms including targeted proteolysis (proteasome) and the removal of signal peptides

LO01 Explain the molecular basis of post-translational modifications, their effects on protein diversity, and their basic biological functions
LO02 It analyzes the regulatory roles of different types of PTMs in intracellular signaling pathways and how they control protein-protein interactions.
LO03 Understands in which amino acid side chains and by which enzymatic processes basic modifications such as phosphorylation, methylation and acetylation (Nα and Nε) occur.
LO04 Interprets the critical effects of modifications such as glycolization and lipidation on protein folding, membrane localization, and stabilization
LO05 Evaluates the role of redox-based changes such as S-nitrosylation and adenylation and oxidation processes in cellular stress and metabolism.
LO06 Describe the enzyme cascades (E1-E2-E3) of ubiquitin and ubiquitin-like modifications and their functions in protein degradation/signal transduction processes.
LO07 It describes the mechanisms of targeted proteolysis (proteasome) and the specific cleavage processes that activate the protein at the molecular level, such as the removal of signaling peptides.
LO08 Analyzes the role of disorders in PTM processes (mutations, misfolding, or enzyme activity changes) in the pathogenesis of cancer, chronic diseases, neurodegenerative diseases, and metabolic disorders at the molecular level; discusses the potential of these modifications as therapeutic targets in a scientific framework using current literature data.
