Course Goal / Objective

To understand the cytological basis of staining techniques for reproductive cells To apply standard and advanced staining protocols for assessing sperm and oocyte quality To interpret stained slides for clinical diagnostics and reproductive research To build laboratory competence in microscopy and sperm evaluation

Course Content

This course provides a comprehensive introduction to classical and modern staining techniques used in the evaluation of sperm and oocyte, embryo and other cells (like granulosa and luteal). It covers the biological significance of each staining method and its application in clinical and research settings. Through a combination of theoretical lectures and hands-on labs, students gain the skills to assess viability, membrane integrity, acrosomal status, DNA fragmentation, and morphology using light and fluorescent microscopy.

Course Learning Outcomes

Order Course Learning Outcomes
LO01 Describe and identify the morphological features of reproductive cells (sperm and oocytes) and explain standard evaluation criteria.
LO02 Apply classical (e.g., Nigrosine-Eosin, Giemsa) and advanced (e.g., HOST, SYBR-PI, TUNEL) staining techniques to assess reproductive cell quality.
LO03 Report semen evaluation results in a scientific format and perform quality control procedures in the reproductive la
LO04 Analyze stained samples using light and fluorescent microscopy to evaluate viability, morphology, membrane integrity, and DNA fragmentation.
LO05 Develop critical decision-making skills based on sperm quality data for use in assisted reproductive technologies (ART).
